From Shinagawa Station, you need to go on Yamamote line (track 2 or 3) to Ebisu Station. At Ebisu Station, you need to change to the Hibiya Line, then travel to Roppongi. If you get out the station towards the Roppongi crossing, you can ask anyone in the area about Midtown. It will be 2 mins walk from Roppongi crossing. Done this route many times & this is the easiest way.
